Description

Principle of the Assay: The Mouse NOPE ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse NOPE in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse NOPE coated on a 96-well plate. Standards and samples are pipetted into the wells and NOPE present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse NOPE antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of NOPE bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: IGDCC4, also known as NOPE, plays essential roles in biologic functions of the cell surface, including cell adhesion, cell migration, and signal transduction. NOPE is identified as an oncofetal marker of murine and human hepatocellular carcinoma (HCC). Its expression is elevated in mouse and human hepatoma cell lines and in mouse hepatomas, but it is not expressed in normal liver or preneoplastic mouse HCC. Furthermore, NOPE is expressed in both AFP -positive and -negative tumors and is specifically expressed in epithelial tumor cells
